Senior Mandrake is the sequel of the film Junior Mandrake. Kuttan (Jagathy Sreekumar) has become enviably rich, and has got rid of the ill-fated statue. Nambiar (Rajan P Dev), a building contractor, is a die-hard atheist. However he is plagued by many a problem in his contracting business, particularly due to the conspiracies plotted by his competitor, Shankara Pillai (Janardhanan). Compelled by his wife Sudharma, who is devoted to God, and his children Pradeep and Sandeep (Jagadeesh and Kalabhavan Navas respectively) Nambiar consents to consult an astrologer Thanku Panikkar (M.S.Thrippunithura) to find if there is any supernatural cause behind their problems. Thanku Asan ascertains that there is an evil spirit residing in the bust of "Junior Mandrake" (in fact a bust in the shape of the antogonist in the movie) kept in their house. Nambiar had been entrusted to handover this bust to someone in the airport in India when he returned from Singapore, but no one had showed up to receive it. Thanku Panikkar prescribes the evil spirit can be got rid of only by presenting the bust to someone who may receive it "with pleasure", who in turn will be taken over by the evil spirit. Nambiar manages to present this bust to his adversary Shankara Pillai on his birthday party, and Thanku Panikkar is apparently proved right as the bad luck of Nambiar is now shifted to Shankara Pillai, also turning Nambiar into an ardent believer in God in the process.

The protagonist of the film is an orphan who takes care of his sister. For an operation of his sister, he needs money. He joins with a known con artist and tries to make money. In one of their joint scams, they were caught red-handed, for not letting them to the handover to the police and for a substantial amount of money they are given a job. To make a wealthy businessman's daughter quit nunnery under the pretense of love. Accidentally the protagonist targets an other woman and makes her to come out of the nunnery. When that women's life is in danger from her step-brother, the protagonist and his companion's joins hands and rescue her and they both rekindle their love.

Jayadeva Sharma hails from an aristocratic Brahmin family. However his family's fortunes disappears and he is fostered by a goldsmith. Eventually he is betrayed by Ananthapadmanabhan, a man who he thinks is Athira's boy friend. To get his money back, he disguises as a sage with the help of his friends Premadasan and gets into the house of Athira's grand father, Kunnamanagalam Neelakantan Vaidyar as a guest. Jayadevan's and Athira's friendship blooms after he saves her from Balagopalan, who is a cousin of her. Premadasan end up falling in love with Vasumathi who is also a cousin of Athira. Near the climax on Athira's wedding day, Jayadevan is held hostage by Ananthapadmanabhan and is saved by Premadasan. At the end Athira's fiancé is caught and Athira and Jayadevan unites.
