The sea Goddess Manimekalai, carries her away from Kaveripoompattinam to the southern island of Manipallavam in Naga Nadu. Manimekalai wakes up from her trance and is mystified at the alien surroundings. While wandering about the island Maṇimekalai comes across the Dharma-seat, the seat on which Buddha had taught and appeased two warring Naga princes, and placed there by the God Indra. Those who worship it miraculously know their previous life. Manimekalai automatically worshiped it and recollects what has happened in her previous life.She comes to a place sanctified by Buddha, and in flash, revelations of her earlier birth dawn on her. She learns that Udaya Kumaran was her husband in her previous birth. Goddess Manimekalai teaches her the art of astral travel and the secret of metamorphosing herself into another being. Goddess Tivatilakai (Deeva-Teelakai or Dvīpa Tilakā) appears now before Manimekalai and inculcates in her the doctrines of Buddhism. Following the instructions of the Goddess, Manimekalai goes around a pond from the middle of which a bowl emerges and places itself on Manaimekalai’s outstretched hands. This is the Amudhasurabhi (cow of abundance), the cornucopia that would never deplete which will always provide food to alleviate hunger.

The film tells the story of twin sisters who get separated at birth due to poverty. One is brought up by a wealthy person, and the other by the mother (Lakshmi). A doctor (Gemini Ganesan) comes to treat the ailing mother, and the mother's daughter falls in love with him, of which the doctor is unaware. On her deathbed, the mother reveals to her daughter the existence of the twin. The young woman goes in search of her sister and by sheer chance, both travel in the same train. The train has an accident and the wealthy sister loses her memory and is presumed dead.

Dharmalingam (M.R. Radha), a rich landlord, is the father of two sons and a daughter. His eldest son has a love marriage with a poor teacher's daughter, Kamala. Dharmalingam disowns them. The son dies and Kamala is left homeless, with a little child. Nathan (MGR), a police officer, gives Kamala shelter in his house and decides to help her fight her case for property. But Kamala is murdered on the way to the court hearing. Fingers point at Dharmalingam and his son Shankar (M.N. Nambhiyar). Shankar and Nathan's sister Uma are in love. Nathan comes to investigate the case and falls in love with Dharmalingam's daughter Sarasu (B. Saroja Devi). Now, Nathan is in a dilemma.
